Targeted editing of OsSWEET13, a bacterial leaf blight susceptible gene in rice using CRISPR tool

Bacterial leaf blight (BLB) of rice is considered to be a disease economic importance as the causes severe yield losses in all growing regions. Transcription activator like effector (TALE) molecules are produced by pathogen, Xanthomonas oryzae pv. (Xoo) bind binding element (EBE) promoter SWEET gene and activates transcription genes, making plant susceptible disease.  Innate resistance Xoo certain genotypes due mutations EBE present upstream regulatory region genes. CRISPR–mediated targeted modification susceptibility gene/promoter an effective approach develop BLB rice. The study was attempt repress TALE triggered signalling via introducing indels OsSWEET13 local popular genotype, CO51 employing CRISPR/Cas9 mediated genome editing tool with view imparting resistance. Agrobacterium-mediated transformation using immature embryos followed regeneration resulted four independent events. Five plants representing three events were found have one nucleotide deletion target sequence.
